Automated platform for yeast cultivation and analysis of secreted phytase in nanoliter droplet arrays
Abstract
We present a method for cultivation of yeast cells and analysis of secreted enzymes in nanoliter (nL) droplets. We combined droplet microfluidics with a spotting platform on top of a fluorescence microscope. Yeast cells are encapsulated in nanoliter volumes using a T-junction and transferred via capillary onto a glass plate with currently up to 10000 hydrophilic spots. Droplets on the plate are post-processed by adding further assay compounds and can be analysed by fluorescence microscopy and mass spectrometry. Show more
